基于混合加权法结合Box-Behnken响应面法优选白及-三七药对提取工艺*

摘要:目的:采用多指标混合加权评分法结合Box-Behnken响应面法(BBD)优选白及-三七药对的提取工艺。方法:利用高效液相色谱(HPLC)法同步检测白及-三七药对中的三七皂苷R1、人参皂苷Rg1、白及苷和人参皂苷Rb1含量,并结合干膏得率作为综合评价指标。在此基础上,运用熵权法-层次分析法(EWM-AHP)混合加权法,确定各指标的权重系数,并开展综合评分分析,通过单因素及BBD法分别考察提取溶剂、液料比、提取时间、提取次数对白及-三七药对提取工艺的影响,优选出最佳的提取工艺。结果:不同提取条件对白及-三七药对指标性成分含量具有一定影响,最佳优化工艺为提取溶剂40%乙醇,料液比为1∶9,提取时间为80 min,提取2次。结论:EWM-AHP混合加权法可用于评价不同提取条件对白及-三七药对的影响,且所得最佳提取工艺具有较好的稳定性与重复性,可为该药对的现代提取研究及复方制剂开发提供参考。

Abstract:
Objective: To optimize the extraction process of Baiji (bletilla striata)-Sanqi (pseudo-ginseng) by multi-index comprehensive weighted scoring method combined with Box-Behnken response surface method (BBD). Methods: High-performance liquid chromatography (HPLC) was used to simultaneously detect the contents of Notoginsenoside R1, ginsenoside Rg1, militarine, ginsenoside Rb1. Notoginsenoside R1, ginsenoside Rg1, militarine, ginsenoside Rb1 and ratio of dry extraction were comprehensive evaluation indicators. On this basis, the entropy weight method analytic hierarchy process (EWM-AHP) mixed weighting method was used to determine the weight coefficients of each indicator, and comprehensive scoring analysis was carried out. The single factor test and BBD were used to investigate the influence of extraction solvent, liquid material ratio, decocting time and decocting times of Baiji (bletilla striata)-Sanqi (pseudo-ginseng) to optimize extraction process parameters. Results: According to the objective weight of each index determined by the mixed weighting method, the comprehensive scoring was performed, and the optimal extraction process condition of Baiji (bletilla striata)- Sanqi (pseudo-ginseng) was determined as adding 9 times of the amount of 40% ethanol, extracting 2 times, each time for 80 minutes. Conclusion: The EWM-AHP mixed weighting method can be used to evaluate the effects of different extraction conditions on the Baiji (bletilla striata)-Sanqi (pseudo-ginseng). The optimized extraction process has been verified to be stable and reproducible, which can provide reference for modern extraction research and development of compound preparation.
