木蝴蝶总黄酮纯化工艺优化及其抗肿瘤活性研究*

摘要:目的:采用大孔吸附树脂分离技术优化木蝴蝶总黄酮纯化工艺并考察其抗肿瘤活性。方法:比较不同型号的大孔吸附树脂对木蝴蝶总黄酮的静态吸附和动态吸附效果,筛选富集效果较好的大孔吸附树脂;结合单因素试验,以总黄酮样品浓度、pH值、最大加样体积、解析溶剂和解析用量为考察因素,优化大孔吸附树脂对木蝴蝶总黄酮的最佳纯化工艺。采用MTT法考察木蝴蝶总黄酮对H1975、HepG2细胞增殖的影响。结果:AB-8大孔吸附树脂的醇解析液中黄酮的纯度最高,木蝴蝶样品质量浓度为6 mg/mL,pH值为2~4,最大加样体积为21 BV时吸附最完全。6 BV体积水洗脱及6 BV体积70%乙醇先后解吸时,总黄酮解析最为完全且纯度最高。MTT试验结果显示,木蝴蝶总黄酮对H1975与HepG2细胞增殖均具有抑制作用,IC50分别为157.7 μg/mL与649.2 μg/mL,表明木蝴蝶总黄酮具有一定的抗肿瘤活性。结论:大孔吸附树脂富集分离木蝴蝶总黄酮的纯化工艺稳定可行,所得的木蝴蝶总黄酮对H1975细胞增殖具有明显抑制作用,但对HepG2细胞增殖抑制作用较弱。

Abstract:Objective: To optimize the purification process of total flavonoids from Muhudie (Oroxylumindicum) using macroporous adsorbent resin separation technique and investigate their antitumour activity. Methods: The static and dynamic adsorption effects of different types of macroporous adsorption resins on the total flavonoids of Muhudie (Oroxylumindicum) were compared to screen the macroporous adsorption resins with better enrichment effects. And then combined with a one-factor test, the concentration of the total flavonoid samples, the pH value, the maximum volume of the sample, the solvent of the resolution, and the amount of the resolution were used as the factors of investigation, to optimize the optimal purification process of the macroporous adsorption resins on the total flavonoids of Muhudie (Oroxylumindicum). In addition, MTT method was used to investigate the effects of total flavonoids of Muhudie (Oroxylumindicum) on the proliferation of H1975 and HepG2 cells. Results: The highest purity of flavonoids was found in the alcohol resolving solution of AB-8 macroporous adsorbent resin, and the most complete adsorption was achieved when the sample concentration of Oroxylumindicum was 6 mg/mL, and the pH value was 2-4, with the maximal volume of 21 BV. The highest purity was achieved when it was eluted with 6 BV volume of water and desorbed with 6 BV volume of 70% ethanol. The results of the MTT assay showed that the total flavonoids of Muhudie (Oroxylumindicum) showed some inhibitory effect on the proliferation of H1975 and HepG2 cells, with IC50 of 157.7 μg/mL and 649.2 μg/mL, respectively, indicating that the total flavonoids in Muhudie (Oroxylumindicum) have certain anti-tumour activity. Conclusion: The purification process for the enrichment and separation of total flavonoids from Muhudie (Oroxylumindicum) by macroporous adsorption resin is stable and feasible, and the obtained total flavonoids from Muhudie (Oroxylumindicum) shows significant inhibitory effect on the proliferation of H1975 cells, but the inhibitory effect on the proliferation of HepG2 cells is weak.
