石见穿多糖通过调控miRNA-145抑制胃癌细胞SGC-7901的增殖、凋亡、侵袭及迁移*

目的:探讨石见穿多糖(SCBP)对胃癌细胞SGC-7901增殖、凋亡、侵袭和迁移的影响及机制。方法:通过MTT和细胞凋亡实验筛选SCBP适宜浓度。将SGC-7901细胞分为正常组(NC组)、SCBP组、miRNA组、SCBP+miRNA组和SCBP+miRNA抑制剂组。采用MTT、流式细胞术、Transwell实验和划痕实验检测细胞增殖率、细胞凋亡率、细胞周期、细胞侵袭能力及细胞迁移能力。Western blotting检测磷脂酰肌醇3-激酶(PI3K)、蛋白激酶B(Akt)、细胞周期蛋白依赖性激酶抑制因子1A(P21)、致癌基因C-MYC(c-Myc)、基质金属蛋白酶-2(MMP-2)及基质金属蛋白酶-9(MMP-9)蛋白表达。结果:SCBP最佳作用浓度为50 μg/mL。SCBP组、miRNA组和SCBP+miRNA组细胞增殖率、侵袭细胞数及伤口愈合率均低于NC组(P<0.05),细胞凋亡率和G1期比例高于NC组(P<0.05)。SCBP+miRNA组细胞增殖率及伤口愈合率低于SCBP组(P<0.05),细胞凋亡率及G1期细胞比例高于SCBP组(P<0.05);SCBP+miRNA抑制剂组细胞增殖率及伤口愈合率高于SCBP组(P<0.01),细胞凋亡率及G1期细胞比例低于SCBP组(P<0.01)。SCBP组、miRNA组和SCBP+miRNA组SGC-7901细胞PI3K、Akt、c-Myc、MMP-2及MMP-9蛋白相对表达量低于NC组(P<0.05),P21蛋白相对表达量高于NC组(P<0.05);SCBP+miRNA组SGC-7901细胞PI3K、Akt、c-Myc、MMP-2及MMP-9蛋白相对表达量低于SCBP组,P21蛋白相对表达量高于SCBP组,差异均有统计学意义(P<0.05);SCBP+miRNA抑制组SGC-7901细胞PI3K、Akt、c-Myc、MMP-2及MMP-9蛋白相对表达量高于SCBP组,P21蛋白相对表达量低于SCBP组,差异均有统计学意义(P<0.01)。结论:SCBP可通过抑制PI3K/Akt/c-Myc/MMP-2/MMP-9信号通路的激活,抑制SGC-7901细胞增殖、凋亡、侵袭及迁移。

Abstract:Objective: To investigate the effects and mechanisms of Salvia Chinensis Benth. Polysaccharides (SCBP) on the proliferation, apoptosis, invasion and migration of SGC-7901 cells. Methods: The suitable SCBP concentration was chosen by MTT and cell apoptosis tests. The SGC-7901 cells were divided into normal group (NC group), SCBP group, miRNA group, SCBP+miRNA group and SCBP+miRNA inhibitor group. The cell proliferation, cell apoptosis, cell cycle, invasion cell number and wound healing rate were evaluated by MTT, flow cytometry, transwell and wound healing assays. Western blotting was used to detect the protein expression of phosphatidylinositide 3-kinases (PI3K), protein kinase B (Akt), cyclin-dependent kinase inhibitor 1A (P21), C-MYC protein (c-Myc), matrix metallopeptidase 2 (MMP-2) and matrix metallopeptidase 9 (MMP-9). Results: The suitable SCBP concentration was 50 μg/mL. The SCBP group, miRNA group, and SCBP+miRNA group showed lower cell proliferation rate, invasion cell number and wound healing rate than NC group (P<0.05), while higher cell apoptosis and G1 phase rate than NC group (P<0.05). SCBP+miRNA group showed lower cell proliferation rate and wound healing rate than SCBP group (P<0.05), while higher cell apoptosis and G1 phase rate than SCBP group (P<0.05). SCBP+miRNA inhibitor group showed higher cell proliferation rate and wound healing rate than SCBP group (P<0.01), while lower cell apoptosis and G1 phase rate than SCBP group (P<0.01). SCBP group, miRNA group, and SCBP+miRNA group showed lower relative expression level of PI3K, Akt, c-Myc, MMP-2 and MMP-9 proteins than NC group (P<0.05), while higher relative expression level of P21 protein than NC group (P<0.05). SCBP+miRNA group showed lower relative expression level of PI3K, Akt,c-Myc, MMP-2 and MMP-9 proteins than SCBP group, while higher relative expression level of P21 protein than SCBP group, with statistically significant differences (P<0.05). SCBP+miRNA inhibitor group showed higher relative expression level of PI3K, Akt, c-Myc, MMP-2 and MMP-9 proteins than SCBP group, while lower relative expression level of P21 protein than SCBP group, with statistically significant differences (P<0.01). Conclusion: SCBP can inhibit the proliferation, apoptosis, invasion and migration of SGC-7901 by inhibition of PI3K/Akt/c-Myc/MMP-2/MMP-9 signaling pathway activation.
