化瘀除痹合剂HPLC多药效成分定量控制及化学计量学联合加权TOPSIS法综合质量评价*

摘要:目的:建立化瘀除痹合剂中多药效成分定量控制的方法,并联合化学计量学及加权优劣解距离(TOPSIS)法评价化瘀除痹合剂的质量。方法:采用高效液相色谱(HPLC)法同时测定16批化瘀除痹合剂中羟基红花黄色素A、红花黄色素A、苦杏仁苷、野黑樱苷、龙胆苦苷、马钱苷酸、獐牙菜苦苷、香附烯酮、α-香附酮、洋川芎内酯H、洋川芎内酯I、洋川芎内酯A和藁本内酯含量,运用统计学软件对16批样品进行差异性分析,挖掘影响化瘀除痹合剂质量的主要因子成分。结果:建立的方法重复性和准确度较好,仪器精密度高,供试品溶液24 h稳定。16批化瘀除痹合剂中13个成分含量存在一定差异。化学计量学分析结果显示16批样品聚为3类,相邻批次聚为一组;龙胆苦苷、羟基红花黄色素A、红花黄色素A、苦杏仁苷、藁本内酯和香附烯酮是重要差异性标志物。加权TOPSIS法结果显示16批样品最优解的欧氏贴近度(Ci)为0.266 7~0.658 7,其中S15的排名靠前,Ci为0.658 7。结论:建立的13个药效成分定量检测的方法操作便捷、结果准确,化学计量学和加权TOPSIS法可评价化瘀除痹合剂的质量。

Abstract:
Objective: To establish a method for quantitative control of multiple bioactive components in Huayu Chubi Mixture (化瘀除痹合剂,HYCBM) and evaluate its quality using chemometrics combined with weighted Technique for Order Preference by Similarity to Ideal Solution (TOPSIS) method. Methods: The contents of 13 bioactive components (hydroxysafflor yellow A, safflor yellow A, amygdalin, prunasin, gentiopicroside, loganic acid, swertiamarin, cyperotundone, α-cyperone, senkyunolide H, senkyunolide I, senkyunolide A, and ligustilide) in 16 batches of HYCBM were simultaneously determined by high-performance liquid chromatography (HPLC). Statistical software was used for differential analysis to identify the main factors affecting the quality of HYCBM. Results: The established method showed good repeatability, accuracy, high instrument precision, and sample stability within 24 h. The contents of the 13 components varied among the 16 batches. Chemometric analysis classified the samples into 3 clusters, with adjacent batches grouped together. Gentiopicroside, hydroxysafflor yellow A, safflor yellow A, amygdalin, ligustilide, and cyperotundone were identified as key markers for quality differentiation. The weighted TOPSIS results showed Euclidean proximity (Ci) values ranging from 0.2667 to 0.6587, with batch S15 ranking highest (Ci=0.6587). Conclusion: The developed method for quantifying 13 bioactive components is convenient and accurate. Chemometrics combined with weighted TOPSIS can effectively evaluate the quality of HYCBM.
