富硒青钱柳-黄精复方抗动脉粥样硬化作用及机制研究*

摘要:

目的:探究富硒青钱柳-黄精复方抗动脉粥样硬化(AS)作用及机制。方法:通过高脂饲料喂养SD大鼠及腹腔注射维生素D3以诱导AS大鼠模型,并灌胃给药富硒青钱柳-黄精复方低、中、高剂量,且阿托伐他汀组为阳性药组。给药8周后,对大鼠体质量、血脂水平进行检测,并观察大鼠主动脉、心脏的组织损伤情况,通过免疫荧光检测大鼠主动脉内皮缩血管肽1endothelin-1ET-1)、内皮型一氧化氮合酶(endothelial nitric oxide synthaseeNOS)、载脂蛋白Eapolipoprotein EApoE)、转化生长因子-β(transforming growth factor-β,TGF-β)TGF-β、细胞间黏附分子-1intercellular cell adhesion molecule-1ICAM-1)、血管细胞黏附分子-1vascular cell adhesion molecule-1VCAM-1)表达情况。结果:连续给药8周后,富硒青钱柳-黄精复方适宜剂量能一定程度改善AS大鼠代谢负担所引起的体质量下降,并调节血脂水平,减少主动脉、心脏的脂质沉积和纤维化症状,有效地保护了AS大鼠的主动脉、心脏。免疫荧光检测结果显示,与模型组比较,富硒青钱柳-黄精复方各剂量组大鼠主动脉组织中eNOSApoETGF-β、ICAM-1VCAM-1表达水平降低,ET-1表达水平升高。结论:富硒青钱柳-黄精复方能够调节AS大鼠血脂水平和脂质代谢紊乱,调控转化生长因子和黏附因子的表达及维持ET-1eNOS平衡,以减缓AS进展。

Abstract:

Objective: To investigate the anti-atherosclerotic effects and underlying mechanisms of the Selenium-enriched Cyclocarya Paliurus-Huangjing (Polygonatum Sibiricum) compound formulation. Methods: SD rats were fed a high-fat diet and received intraperitoneal injections of vitamin D3 to induce an atherosclerosis (AS) model. The rats were then administered low, medium and high doses of the Selenium-enriched Cyclocarya Paliurus-Huangjing (Polygonatum Sibiricum) compound formulation via oral gavage, with atorvastatin group serving as the positive control group. After 8 weeks of treatment, body weight and blood lipid levels were measured. Additionally, tissue damage in the aorta and heart was assessed. Immunofluorescence was used to detect the expression of endothelin-1 (ET-1), endothelial nitric oxide synthase (eNOS), apolipoprotein E (ApoE), transforming growth factor-β (TGF-β), intercellular cell adhesion molecule-1 (ICAM-1), and vascular cell adhesion molecule-1 (VCAM-1) in the aorta. Results: After 8 weeks of continuous administration, appropriate doses of the Selenium-enriched Cyclocarya Paliurus-Huangjing (Polygonatum Sibiricum) compound formulation significantly mitigated the body weight loss induced by metabolic burden in AS rats. The formulation also regulated blood lipid levels, and reduced lipid deposition and fibrosis in the aorta and heart, effectively protecting these tissues in AS rats. Immunofluorescence results showed that compared to the model group, the expression levels of eNOS, ApoE, TGF-β, ICAM-1 and VCAM-1 in the aortic tissue of the each dose group of Selenium-enriched Cyclocarya Paliurus-Huangjing (Polygonatum Sibiricum) compound formulation groups were decreased, while the expression level of ET-1 was increased. Conclusion: The Selenium-enriched Cyclocarya Paliurus-Huangjing (Polygonatum Sibiricum) compound formulation can regulate blood lipid levels and lipid metabolism disorders, modulate the expression of transforming growth factors and adhesion molecules, and maintain the balance between ET-1 and eNOS, thereby slowing the progression of AS in AS rat.
